Tirzepatide is both a GLP-1 agonist AND a GIP agonist, blocking both receptors. Semaglutide is only a GLP-1 agonist. Tirzepatide, thanks to its dual agonist mechanism of action, showed greater weight loss in clinical trials than Semaglutide- 22.5% (Tirzepatide) vs 10-14% (Semaglutide). Anecdotally, patients report significantly less side effects with Tirzepatide leading to increased drug tolerability and follow through with treatment and results.
